© 2015 Elsevier Inc.Toxoplasma gondii is a protozoan pathogen in the phylum Apicomplexa that resides within an intracellular parasitophorous vacuole that is selectively permeable to small molecules through unidentified mechanisms. We have identified GRA17 as a Toxoplasma-secreted protein that localizes to the parasitophorous vacuole membrane and mediates passive transport of small molecules across the PVM. GRA17 is related to the putative Plasmodium translocon protein EXP2 and conserved across PV-residing Apicomplexa. The PVs of GRA17-deficient parasites have aberrant morphology, reduced permeability to (...) small molecules, and structural instability. GRA17-deficient parasites proliferate slowly and are avirulent in mice. These GRA17-deficient phenotypes are rescued by complementation with Plasmodium EXP2. GRA17 functions synergistically with a related protein, GRA23. Exogenous expression of GRA17 or GRA23 alters the membrane conductance properties of Xenopus oocytes in a manner consistent with a large non-selective pore. Thus, GRA17 and GRA23 provide a molecular basis for PVM permeability and nutrient access. (shrink)

The comorbidity of chronic pain and opioid addiction is a serious problem that has been growing with the practice of prescribing opioids for chronic pain. Neuroimaging research has shown that chronic pain and opioid dependence both affect brain structure and function, but this is the first study to evaluate the neurophysiological alterations in patients with comorbid chronic pain and addiction. Eighteen participants with chronic low back pain and opioid addiction were compared with eighteen age- and sex-matched healthy individuals in a (...) pain-induction fMRI task. Unified structural equation modeling (SEM) with Lagrange multiplier (LM) testing yielded a network model of pain processing for patient and control groups based on 19 a priori defined regions. Tests of differences between groups on specific regression parameters were determined on a path-by-path basis using z-tests corrected for the number of comparisons. Patients with the chronic pain and addiction comorbidity had increased connection strengths; many of these connections were interhemispheric and spanned regions involved in sensory, affective, and cognitive processes. The affected regions included those that are commonly altered in chronic pain or addiction alone, indicating that this comorbidity manifests with neurological symptoms of both disorders. Understanding the neural mechanisms involved in the comorbidity is crucial to finding a comprehensive treatment, rather than treating the symptoms individually. (shrink)

In April 2016, Daniela Frauchiger and Renato Renner published an article online in which they introduce a Gedankenexperiment that led them to conclude that single-world interpretations of quantum theory cannot be self-consistent. In a new version of the paper, published in September 2018, the authors moderate their original claim by concluding that quantum theory cannot be extrapolated to complex systems, at least not in a straightforward manner. The purpose of this short article is to clarify the core of the F-R (...) argument, in order to show how the contradiction is obtained. On the basis of this clarification, we argue that the result of the F-R argument has been overestimated and should be reconsidered from a more cautious perspective. (shrink)

En el presente artículo nos proponemos realizar una comparación de la posición de D’Agostino con la de otro de los representantes del convencionalismo: William Morgan. A D’Agostino nos referiremos en el primer apartado y a Morgan en el segundo. Nos preguntaremos si Morgan, además de inscribir sus tesis dentro del convencionalismo, participa al mismo tiempo de la concepción internalista del deporte. In this paper we propose a comparison of the position of D’Agostino with other rpresentatives of convention: Wilüam Morgan. We refer to D’Agostino’s position in the first section and in the second section to Morgan ’s. We ask whether Morgan, moreover of register his thesis within the Conventionalism, participaos also of the internaüst conception of sport. Although both authors are conventionalists, there are many different shades that fit between their positions be highüghted, in order to clarify its proximity to other phihsophical theories of port. En este artículo presento de forma crítica la propuesta de ética del deporte como «bolsa de virtudes» que elaboran tres autores estadounidenses que son Lumpkin, Stoll y Beller. Dicha propuesta debe entenderse como una manifestación de sustancialismo neoaristotélico en deporte, con una clara influencia de MacIntyre. Mi objetivo es mostrar cómo dicho sustancialismo no es suficiente para una propuesta ética del deporte, por lo que abogo por incorporar algunos de los presupuestos del procedimentalismo ético aplicado al deporte. In this article (...) the author presents critically the proposed sports ethics as «bag of virtues» that Americans made three authors are Lumpkin, Stoll and Beller. This proposal should be seen as a manifestation of substantialism neoaristhotelic in sport, with a clear influence of MacIntyre. Indeed, the technological and cultural advancement of the species has been widely acknowledged to rest upon human domination and control of nature. Yet, by the 1960s, the (...) idea of culture in confrontation with nature was being challenged by science, philosophy and the environmental movement. Anthropology is increasingly concerned with such issues as they become more urgent for humankind as a whole. Reading the Republic without reference to the less familiar Laws can lead to a distorted view of Plato's political theory. In the Republic the philosopher describes his ideal city; in his last and longest work he deals with the more detailed considerations involved in setting up a second-best 'practical utopia.' The relative neglect of the Laws has stemmed largely from the obscurity of its style and the apparent chaos of its organization so that, although good translations now exist, students of (...) philosophy and political science still find the text inaccessible. This first full-length philosophical introduction to the Laws will therefore prove invaluable. The opening chapters describe the general character of the dialogue and set it in the context of Plato's political philosophy as a whole. Each of the remaining chapters deals with a single topic, ranging over material scattered through the text and so drawing together the threads of the argument in a stimulating and readily comprehensible way. Those topics include education, punishment, responsibility, religion, virtue and pleasure as well as political matters and law itself. Throughout, the author encourages the reader to think critically about Plato's ideas and to see their relevance to present-day philosophical debate. No knowledge of Greek is required and only a limited background in philosophy. Although aimed primarily at students, the book will also be of interest to more advanced readers since it provides for the first time a philosophical, as opposed to linguistic or historical, commentary on the Laws in English. HUME’S DISCUSSION OF THE EVIDENCE FOR MIRACLES RELATES TO THIS KIND AND IS INTERPRETABLE AS AN ARGUMENT AGAINST ITS POSSIBILITY. ALSO THERE IS AN ARGUMENT THAT THE EXPRESSION "VIOLATION OF NATURAL LAW" SIGNIFIES A CONFUSION IN WHICH THE IDEAS OF NATURAL LAW AND LEGAL LAW COLLAPSE INTO EACH OTHER. NEITHER OF (...) THESE ARGUMENTS IS EFFICACIOUS. FURTHERMORE, THE CONTENTION THAT THERE CAN BE NO SUCH THING AS ESTABLISHING THE "ABSENCE" OF A NATURAL CAUSE IS OPEN TO OBJECTION.
